Description

  • Pyrrolo-pyrrole group of NSAID
  • Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
  • Inhibition of prostaglandin synthesis
  • Biologically active S-form
  • Linear pharmacokinetic property
  • Highly protein bound
  • Mainly metabolized in liver
  • Metabolites and unchanged drug excretion in urine

Pharmacology

  • Potent analgesic NSAID
  • Inhibits cyclooxygenase enzyme system and prostaglandin synthesis
  • Minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

  • Tablet: Recommended dose is 10 mg every 4-6 hours for up to 7 days
  • Injection: Single or multiple doses for acute pain management with specific dosing guidelines for different age groups and conditions
  • Eye Drops: Recommended dosing regimen for adults

Interaction

  • Increased side effects with other NSAIDs or aspirin
  • Enhanced anti-coagulant effect with anti-coagulants
  • Reduced anti-hypertensive effect with Beta Blockers
  • Increased risk of renal impairment with ACE Inhibitors
  • Enhanced toxicity of methotrexate

Contraindications

  • Hypersensitivity to Ketorolac or other NSAIDs
  • Not for use in children under 16 years
  • Contraindicated as prophylactic analgesic before surgery
