Description
- Pyrrolo-pyrrole group of NSAID
- Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibition of prostaglandin synthesis
- Biologically active S-form
- Linear pharmacokinetic property
- Highly protein bound
- Mainly metabolized in liver
- Metabolites and unchanged drug excretion in urine
Pharmacology
- Potent analgesic NSAID
- Inhibits cyclooxygenase enzyme system and prostaglandin synthesis
- Minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet: Recommended dose is 10 mg every 4-6 hours for up to 7 days
- Injection: Single or multiple doses for acute pain management with specific dosing guidelines for different age groups and conditions
- Eye Drops: Recommended dosing regimen for adults
Interaction
- Increased side effects with other NSAIDs or aspirin
- Enhanced anti-coagulant effect with anti-coagulants
- Reduced anti-hypertensive effect with Beta Blockers
- Increased risk of renal impairment with ACE Inhibitors
- Enhanced toxicity of methotrexate
Contraindications
- Hypersensitivity to Ketorolac or other NSAIDs
- Not for use in children under 16 years
- Contraindicated as prophylactic analgesic before surgery
