Pharmacological Action
Dopamine Antagonist, Gastroprokinetic
Mode of Action
Blocks dopamine receptors in Chemoreceptor Trigger Zone (CTZ) and stomach, restores normal motility and tone of upper GI tract
Gastrointestinal Effects
- Facilitates Gastric Emptying
- Enhances Antral and Duodenal Peristalsis
- Regulates Contraction of the Pylorus
- Increases Esophageal Peristalsis
- Regulates Lower Esophageal Sphincter Pressure
Usability
- Taken before meals and bedtime
- Relieves Nausea, Vomiting, Indigestion
Dosage Recommendations
- Adults: 10-20 mg, every 6-8 hours daily
- Children: 2-4 ml suspension/10 kg body weight, every 6-8 hours daily

80 mg
Interaction
- Contraindicated with anticholinergic drugs
- Simultaneous use with antacids and antisecretory drugs lowers oral bioavailability
- Metabolized through CYP3A4 pathway
Contraindications
- Known hypersensitivity
- Neonates
- Gastrointestinal hemorrhage
- Mechanical obstruction or perforation
- Prolactin releasing pituitary tumor (prolactinoma)
Side Effects
- Transient intestinal cramps
- Rare extrapyramidal phenomena
