Indications
- Treatment of urinary tract infection
- Caused by susceptible Gram-negative organisms
- Including Proteus species
- Klebsiella species
- Enterobacter species
- Escherichia coli
Pharmacology
- Synthetic narrow spectrum antibacterial
- Bacteriostatic or bactericidal
- Inhibits bacterial DNA synthesis
- Rapidly and completely absorbed from the G.I. Tract
- Distributed to most tissues, especially to the kidney and urine
Dosage & Administration
- Adult dosage: 1g every 6 hours for 7 days, then reducing to 500mg every 6 hours
- Children dosage: 13.75mg per kg body weight every six hours for one or two weeks, as prescribed by the physician
Interaction
- Concomitant use with melphalan can cause severe blood-containing diarrhea
- Probenecid inhibits tubular secretion of Nalidixin
- Chlorpromazine and Perphenazine may potentiate the effect of Nalidixin
Contraindications
- Infants under 3 months
- Epilepsy
- CNS lesions
Side Effects
- Gastro-intestinal disturbances including nausea, vomiting, diarrhea
- Haemolysis in G6PD deficiency
- Allergic reactions including urticaria, rashes, fever, arthralgia, eosinophilia
- Myalgia, muscle weakness, phototoxicity, jaundice, visual disturbances, convulsions
Pregnancy & Lactation
- Possibility of causing cartilage and DNA damage
- Excreted in breast milk
- Report of hemolytic anemia in a breastfed child of an azotemic mother
Precautions & Warnings
- Risk-benefit consideration during first trimester of pregnancy and breastfeeding
- Imapired renal or hepatic function
