Pharmacology
Blocks dopamine receptors in Chemoreceptor Trigger Zone and stomach. Restores normal gastrointestinal motility and tone. Facilitates gastric emptying and enhances peristalsis. Increases esophageal peristalsis and lower esophageal sphincter pressure.
Dosage & Administration
- Adult dose: 10-20 mg every 6-8 hours daily
- Children's dose: 0.2-0.4 mg/kg body weight every 6-8 hours daily
- Rectal administration for adults and children
Interaction
- Antagonism with anticholinergic drugs
- Decreased bioavailability with antacids and antisecretory drugs
- Metabolic pathway through CYP3A4
- Potential inhibition with CYP3A4 inhibitors
- No potentiation of neuroleptics
