Pharmacology
Domperidone is a dopamine antagonist that blocks dopamine receptors in the Chemoreceptor Trigger Zone (CTZ) and stomach.
Dosage & Administration
- General Dosage:
- Adults: 10-20 mg every 6-8 hours daily
- Children: 2-4 ml/10 kg body weight every 6-8 hours daily
- Specific Conditions:
- Dyspeptic Symptom: 10-20 mg every 6-8 hours daily
- Acute and Sub-Acute Conditions: 20 mg every 6-8 hours daily
- Rectal Administration:
- Adults (including elderly): 30-60 mg every 4-8 hours
- Children: Max daily dose of 30 mg for 10-25 kg weight
Interaction
- Concomitant Administration: Anticholinergic drugs may antagonize the antidyspeptic effect.

- Antacids
- Antisecretory drugs
- CYP3A4 inhibitors

Contraindications
- Known hypersensitivity to the drug
- Neonates
- Gastrointestinal hemorrhage
- Mechanical obstruction or perforation
- Prolactin releasing pituitary tumor (prolactinoma)
Side Effects
- Transient intestinal cramps
- Extrapyramidal phenomena
- Hyperprolactinaemia
- Rare allergic reactions
Pregnancy & Lactation
- Animal Studies: Doses up to 160 mg/kg/day did not produce teratogenic effects.
- Human Use: Use during the first trimester if justified by therapeutic benefit. Caution in lactating mothers.
Precautions & Warnings
- Caution in children due to risk of extrapyramidal reactions
- Caution in hepatic impairment

- Caution in liver disorders
- Caution in kidney disorders
Overdose Effects
Symptoms may include drowsiness, disorientation, and extrapyramidal reactions. Management with activated charcoal and close observation.
