Description

A dopamine antagonist medication used to treat nausea and vomiting

DosageAndAdministration

  • Should be taken 15-30 minutes before meals
  • Usual recommended oral dose for adults: 10-20 mg every 6-8 hours daily
  • Maximum dose for adults: 80 mg daily
  • Recommended dose for children based on body weight

Indications

  • Treatment of dyspeptic symptom complex
  • Acute nausea and vomiting of various origins
  • Parkinson's disease-induced nausea and vomiting
  • Speeding barium transit in radiological studies

Pharmacology

Acts as a dopamine antagonist, mainly blocking dopamine receptors in the Chemoreceptor Trigger Zone and stomach. Restores normal motility and tone of the upper gastrointestinal tract, facilitates gastric emptying, enhances peristalsis, etc.

Interaction

  • Anticholinergic drugs may antagonize the antidyspeptic effect
  • Concomitant use with CYP3A4 inhibitors may increase plasma levels of domperidone

Contraindications

  • Known hypersensitivity to the drug
  • Neonates
  • Gastrointestinal hemorrhage, obstruction, or perforation
  • Prolactin releasing pituitary tumor

SideEffects

  • Rare side effects include transient intestinal cramps, extrapyramidal phenomena, hyperprolactinaemia
  • Rare allergic reactions such as rash and urticaria

PregnancyAndLactation

  • Should only be used during the first trimester if justified by therapeutic benefit
  • Excreted in breast milk, caution advised for nursing mothers
