Indications

  • Prophylactic treatment of bronchial asthma
  • Symptomatic treatment of allergic conditions including rhinitis and conjunctivitis
  • Alleviating complications of itching, pain, and tenderness associated with neurofibroma
  • Symptomatic treatment of allergy such as hayfever, urticaria

Pharmacology

  • Anti-allergic properties
  • Antihistamine properties
  • Anti-anaphylactic properties
  • Sustained inhibitory effect on histamine reactions

Dosage & Administration

  • Adult dosage: 1 mg twice daily with food
  • Increased dosage to 2 mg twice daily in severe cases
  • Children dosage: 1 mg twice daily with food
  • Elderly dosage: Same as adult dose or as advised by the physician

Interaction

  • Potentiation of sedatives, hypnotics, antihistamines, and alcohols
  • Reversible fall in platelet count with concomitant use of oral antidiabetic agents
  • Avoidance of combination with certain medications

Contraindications

  • Avoidance of combination with oral antidiabetic agents during reversible fall in platelet count
  • Caution in use during pregnancy and breastfeeding

Side Effects

  • Drowsiness
  • Dry mouth
  • Slight dizziness

Pregnancy & Lactation

  • Not recommended for use during pregnancy and lactation
  • No evidence of teratogenic effect

Precautions & Warnings

  • Continuation of previous treatment for a minimum of two weeks after starting Orotifen
  • Special caution with systemic corticosteroids and ACTH
  • Impaired reactions on first day of treatment
  • Avoidance of alcoholic drinks
  • Potentiation of effects of sedatives, hypnotics, antihistamines, and alcohol

Overdose Effects

  • Confusion
  • Drowsiness
  • Headache
  • Bradycardia
  • Respiratory depression
  • Recommendation for gastric lavage or emesis
  • General supportive treatment
