Pharmacology
- Dopamine antagonist
- Blocks dopamine receptors in the Chemoreceptor Trigger Zone and stomach
- Gastroprokinetic action
- Restores normal motility and tone of the upper gastrointestinal tract
- Facilitates gastric emptying
- Increases esophageal peristalsis
- Regulates contraction of the pylorus
DosageAdministration
- Should be taken 15-30 minutes before meals
- Adults: 10-20 mg every 6-8 hours daily
- Children: 2-4 ml suspension/10 kg body weight every 6-8 hours daily
- Maximum dose for adults: 80 mg daily
- Maximum period of treatment for acute nausea and vomiting: 12 weeks
Interaction
- Antagonized by anticholinergic drugs
- Lowered oral bioavailability when given simultaneously with antacids and antisecretory drugs
- Metabolized through CYP3A4
- Inhibited by CYP3A4 inhibitors such as azole antifungals and macrolide antibiotics
- May influence the absorption of orally administered drugs
Contraindications
- Known hypersensitivity to the drug
- Neonates
- Gastrointestinal hemorrhage
- Mechanical obstruction or perforation
- Prolactin releasing pituitary tumor (prolactinoma)
