Description

Dosage & Administration

Should be taken 15-30 minutes before meals, maximum oral dose for adults is 80 mg daily

Pharmacology

Dopamine antagonist that blocks dopamine receptors in Chemoreceptor Trigger Zone (CTZ) and stomach

Interaction

  • Antagonizes antidyspeptic effect of anticholinergic drugs
  • Lower oral bioavailability when given simultaneously with antacids and antisecretory drugs
  • Inhibited by CYP3A4 inhibitors like azole antifungals, macrolide antibiotics, HIV protease inhibitors, nefazodone
  • Could influence absorption of concomitantly orally administered drugs

Contraindications

  • Known hypersensitivity to the drug
  • Neonates
  • Gastrointestinal hemorrhage, mechanical obstruction or perforation
  • Prolactin releasing pituitary tumor (prolactinoma)

Side Effects

  • Transient intestinal cramps
  • Rare extrapyramidal phenomena
  • Increase in plasma prolactin level
  • Rare allergic reactions like rash and urticaria

Pregnancy & Lactation

Should be used with caution during the first trimester of pregnancy, excreted in breast milk
