Description
- Dopamine antagonist medication
- Used to treat dyspeptic symptom complex, acute nausea and vomiting, Parkinson's disease, and speeding barium transit in radiological studies

Dosage & Administration
Should be taken 15-30 minutes before meals, maximum oral dose for adults is 80 mg daily

Pharmacology
Dopamine antagonist that blocks dopamine receptors in Chemoreceptor Trigger Zone (CTZ) and stomach
Interaction
- Antagonizes antidyspeptic effect of anticholinergic drugs
- Lower oral bioavailability when given simultaneously with antacids and antisecretory drugs
- Inhibited by CYP3A4 inhibitors like azole antifungals, macrolide antibiotics, HIV protease inhibitors, nefazodone
- Could influence absorption of concomitantly orally administered drugs
Contraindications
- Known hypersensitivity to the drug
- Neonates
- Gastrointestinal hemorrhage, mechanical obstruction or perforation
- Prolactin releasing pituitary tumor (prolactinoma)
Side Effects
- Transient intestinal cramps
- Rare extrapyramidal phenomena
- Increase in plasma prolactin level
- Rare allergic reactions like rash and urticaria
Pregnancy & Lactation
Should be used with caution during the first trimester of pregnancy, excreted in breast milk
