Indications
- Dyspeptic symptom complex including epigastric fullness, abdominal pain, nausea, and vomiting
- Acute nausea and vomiting
- Parkinson's disease-related nausea and vomiting
- Speeding barium transit in radiological studies
Pharmacology
- Dopamine antagonist action
- Blockage of dopamine receptors in CTZ and stomach
- Gastroprokinetic action
- Enhancement of gastric emptying and intestinal peristalsis
Dosage & Administration
- Adults:
- 10-20 mg, every 6-8 hours daily
- Maximum dose of 80 mg daily
- Children:
- 2-4 ml suspension/10 kg body weight
- 0.4-0.8 ml paediatric drops/10 kg body weight
Interactions
- Antagonism with anticholinergic drugs
- Lowered oral bioavailability with antacids and antisecretory drugs
- Metabolic pathway through CYP3A4
- Potential interaction with azole antifungals and macrolide antibiotics
Contraindications
- Known hypersensitivity to domperidone
- Neonates
- Gastrointestinal hemorrhage, mechanical obstruction, or perforation
- Prolactin releasing pituitary tumor
