Indications

  • Dyspeptic symptom complex including epigastric fullness, abdominal pain, nausea, and vomiting

Pharmacology

  • Dopamine antagonist action
  • Blockage of dopamine receptors in CTZ and stomach
  • Gastroprokinetic action

Dosage & Administration

  • Adults:
    • 10-20 mg, every 6-8 hours daily
    • Maximum dose of 80 mg daily
  • Children:
    • 2-4 ml suspension/10 kg body weight
    • 0.4-0.8 ml paediatric drops/10 kg body weight

Interactions

  • Antagonism with anticholinergic drugs
  • Lowered oral bioavailability with antacids and antisecretory drugs
  • Metabolic pathway through CYP3A4
  • Potential interaction with azole antifungals and macrolide antibiotics

Contraindications

  • Known hypersensitivity to domperidone
  • Neonates
  • Gastrointestinal hemorrhage, mechanical obstruction, or perforation
  • Prolactin releasing pituitary tumor
