Description

  • Doxair is used to treat bronchial asthma, bronchospasm, chronic obstructive pulmonary disease (COPD), and pulmonary disease with spastic bronchial component.
  • Doxophylline is a bronchodilator that selectively inhibits phosphodiesterase 4, relaxes bronchial smooth muscle, and has decreased affinities toward adenosine A1 and A2 receptors.
  • Dosage for elderly: 200 mg tablet two or three times daily, adults: 400 mg tablet two or three times daily, children >12 years: 10 ml syrup or 200 mg tablet two or three times daily.
  • Doxair should not be administered with other xanthine preparations and caution should be exercised with ephedrine or other sympathomimetic drugs.
  • Doxophylline is contraindicated in individuals with known hypersensitivity to the drug or other xanthine derivatives, acute myocardial infarction, hypotension, and during lactation.

Pharmacology

Dosage & Administration

  • Elderly: 200 mg tablet two or three times daily
  • Adults: 400 mg tablet two or three times daily
  • Children >12 years: 10 ml syrup or 200 mg tablet two or three times daily
  • Children 6-12 years: 6-9 mg/kg body weight two times daily

Interactions

  • Not to be administered with other xanthine preparations
  • Caution with ephedrine or other sympathomimetic drugs
  • Concurrent administration with drugs like erythromycin, TAO, lincomycin, clindamycin, allopurinol, cimetidine, influenza vaccine, and propranolol may reduce hepatic clearance
  • Phenytoin, other anticonvulsants, and cigarette smoking may increase clearance

Contraindications

  • Known hypersensitivity to the drug or other xanthine derivatives
  • Acute myocardial infarction
  • Hypotension
  • During lactation

  • Insomnia
  • Tachycardia
  • Extrasystoles
  • Tachypnea
  • Rare cases of hyperglycemia or albuminuria
  • Severe cardiac arrhythmias and tonic-clonic seizure in case of overdose

Pregnancy & Lactation

  • Animal tests show no interference with pre and postnatal growth
  • Use during pregnancy to be evaluated carefully on risk-benefit ratio basis
  • Contraindicated during lactation
  • No effects on ability to drive and use machines

Precautions & Warnings

  • Factors reducing hepatic clearance may increase plasma levels
  • Caution in patients with cardiac disease, hypertension, severe hypoxemia, hyperthyroidism, chronic corpulmonale, congestive heart failure, liver disease, peptic ulcer, and renal impairment
  • No risk of addiction or dependence

Overdose Effects

  • Severe cardiac arrhythmias
  • Tonic-clonic seizure
  • No specific antidote
