Indications

• Control of clinical spasticity
• Manifestations of upper motor neuronal disorders
• Chronic spasticity resulting from spinal cord injury
• Chronic spasticity resulting from stroke
• Chronic spasticity resulting from cerebral palsy
• Chronic spasticity resulting from multiple sclerosis
• Restoration of residual function
• Malignant hyperthermia prevention
• Malignant hyperthermia recurrence prevention

Pharmacology

• Relaxation of skeletal muscle
• Affects contractile response of muscle
• Interference with release of Ca++ from sarcoplasmic reticulum

Dosage & Administration

• Gradual titration schedule for adults
• Gradual titration schedule for paediatric patients
• Maximum accepted dosage
• Preoperative dosage for malignant hyperthermia
• Post-crisis follow-up dosage for malignant hyperthermia

Interaction

• Drowsiness with therapy
• Concomitant administration of CNS depressants
• Hepatotoxicity in women over 35 years with estrogen therapy

Contraindications

• Active hepatic disease

Pregnancy & Lactation

• Pregnancy Category C
• Use only if potential benefit justifies potential risk to fetus
• Not for use in nursing mothers

• Potential for liver damage in long-term use
• Caution against driving or hazardous occupations
• Exercise caution in concomitant administration of tranquilizing agents
• Possibility of photosensitivity reaction
• Recognition of fatal and non-fatal liver disorders
• Baseline liver function studies

• Return to normal laboratory values in some patients
