Indications
- Treatment of premature ejaculation in men 18-64 years of age
- Persistent or recurrent ejaculation with minimal sexual stimulation
- Marked personal distress or interpersonal difficulty as a consequence of PE
- Poor control over ejaculation
Pharmacology
- Inhibition of neuronal reuptake of serotonin
- Potentiation of neurotransmitter's action at pre- and post-synaptic receptors
- Rapid absorption with maximum plasma concentrations occurring 1-2 hours after intake
- Absolute bioavailability of 42%
- Extensive metabolism to multiple metabolites
- Rapid distribution with a mean steady state volume of distribution of 162 L
- Terminal half-life of approximately 19 hours
Dosage & Administration
- Recommended starting dose of 30 mg for adult men
- Maximum recommended dosing frequency of once every 24 hours
- Not recommended for use in individuals below 18 years of age
- Caution advised in patients with mild or moderate renal impairment
- Contraindicated in patients with moderate and severe hepatic impairment
Interaction
- Caution advised if concomitant administration with CNS active medicinal products is required
- Prescribed with caution in patients who use PDE5 inhibitors
- Prescribed with caution in patients who use alpha adrenergic receptor antagonists
- Caution advised when used in patients taking Warfarin chronically
- Concomitant use of alcohol could increase the chance or severity of adverse reactions
Contraindications
- Known hypersensitivity to Dapoxetine Hydrochloride
- Significant pathological cardiac conditions
- Concomitant treatment with monoamine oxidase inhibitors (MAOIs), thioridazine
- Concomitant treatment with serotonin reuptake inhibitors, or other medicinal/herbal products with serotonergic effects
Side Effects

- Headache
- Diarrhea
- Insomnia
- Low rates of sexual dysfunction
- Very mild adverse effects on loss of libido and ED
Pregnancy & Lactation
- Not indicated for use by women
- Excretion through human breast milk unknown
Precautions & Warnings
- Not to be used in combination with recreational drugs
- Possible serious reactions if combined with serotonergic recreational drugs
- Combining alcohol with Dapoxen may increase alcohol-related neurocognitive effects
- Possibility of experiencing syncope
- An orthostatic test should be performed before initiating therapy
- Reports of bleeding abnormalities with SSRIs
- Caution advised in concomitant use with medicinal products known to affect platelet function
Overdose Effects
- No reported case of overdose
- Symptoms of overdose with SSRIs include serotonin-mediated adverse reactions such as somnolence, gastrointestinal disturbances, tachycardia, tremor, agitation, and dizziness
- No evidence of withdrawal syndrome
