Indications
- Treatment of premature ejaculation (PE) in men 18 to 64 years of age
- Persistent or recurrent ejaculation with minimal sexual stimulation before, on, or shortly after penetration
- Marked personal distress or interpersonal difficulty as a consequence of PE
- Poor control over ejaculation
Pharmacology
- Inhibition of neuronal reuptake of serotonin
- Potentiation of the neurotransmitter's action at pre-and post synaptic receptors
- Rapid absorption with maximum plasma concentrations occurring approximately 1-2 hours after tablet intake
- Extensive metabolism to multiple metabolites primarily through biotransformational pathways
- Rapid elimination with a terminal half-life of approximately 19 hours
Dosage & Administration
- Recommended starting dose for adult men is 30 mg, taken as needed approximately 1 to 3 hours prior to sexual activity
- Maximum recommended dosing frequency is once every 24 hours
- Not recommended for use in individuals below 18 years of age
- Caution advised in patients with mild or moderate renal impairment
- Contraindicated in patients with moderate and severe hepatic impairment
Interaction
- Caution advised if concomitant administration of Dejac and CNS active medicinal products is required
- Prescribed with caution in patients who use PDE5 inhibitors and alpha adrenergic receptor antagonists
- Caution advised when used in patients taking Warfarin chronically
- Concomitant use of alcohol and Dejac could increase the chance or severity of adverse reactions
Contraindications
- Known hypersensitivity to Dapoxetine Hydrochloride
- Significant pathological cardiac conditions
- Concomitant treatment with monoamine oxidase inhibitors (MAOIs), thioridazine
- Concomitant treatment with serotonin reuptake inhibitors or other medicinal/herbal products with serotonergic effects

Pregnancy & Lactation
- Not indicated for use by women
- Unknown excretion through human breast milk
Precautions & Warnings
- Patients should be advised not to use Dejac in combination with recreational drugs
- Use of Dejac with recreational drugs with serotonergic activity may lead to potentially serious reactions
- Patients should be advised to avoid alcohol while taking Dejac
- Orthostatic test should be performed before initiating therapy
- Caution is advised in patients taking Dejac with medicinal products known to affect platelet function
Overdose Effects
- No case of overdose reported
- Symptoms of overdose with SSRIs include serotonin-mediated adverse reactions such as somnolence, nausea, vomiting, tachycardia, tremor, agitation, and dizziness
- No evidence of withdrawal syndrome observed in clinical trial for 62 days of daily or as needed dosing with 60 mg Dejac
