Indications
- Treatment of premature ejaculation (PE) in men 18 to 64 years of age
- Persistent or recurrent ejaculation with minimal sexual stimulation
- Marked personal distress or interpersonal difficulty
- Poor control over ejaculation
Pharmacology
- Inhibition of neuronal reuptake of serotonin
- Potentiation of the neurotransmitter's action
- Rapid absorption with maximum plasma concentrations occurring 1-2 hours after tablet intake
- Absolute bioavailability of 42%
- Extensive metabolization to multiple metabolites
- Rapid elimination with a half life of approximately 19 hours
Dosage & Administration
- Recommended starting dose for adult men is 30 mg
- Maximum recommended dosing frequency is once every 24 hours
- Dapoxetine may be taken with or without food
- Not recommended for use in individuals below 18 years of age
- Caution advised in patients with renal or hepatic impairment
Interactions
- Caution advised with CNS active medicinal products
- Prescribed with caution in patients who use PDE5 inhibitors or alpha adrenergic receptor antagonists
- Caution is advised when used in patients taking Warfarin chronically
- Concomitant use of alcohol and Plezer could increase the chance or severity of adverse reactions
Contraindications
- Known hypersensitivity to Dapoxetine Hydrochloride
- Significant pathological cardiac conditions
- Concomitant treatment with MAOIs, thioridazine, or within 14 days of discontinuing treatment with MAOIs, thioridazine
- Concomitant treatment with serotonin reuptake inhibitors or other medicinal/herbal products with serotonergic effects

Pregnancy & Lactation
- Not indicated for use by women
- Not known if excreted through human breast milk
Precautions & Warnings
- Patients advised not to use Plezer in combination with recreational drugs
- Possibility of syncope, orthostatic hypotension, and haemorrhage
- Caution advised in concomitant use with medicinal products known to affect platelet function or anticoagulants
- Plezer should be prescribed with caution in patients who use alpha adrenergic receptor antagonists
Overdose Effects
- No case of overdose reported
- Symptoms of overdose with SSRIs include serotonin-mediated adverse reactions
