Indications
- Treatment of premature ejaculation in men 18 to 64 years of age
- Persistent or recurrent ejaculation with minimal sexual stimulation
- Marked personal distress or interpersonal difficulty as a consequence of PE
- Poor control over ejaculation
Pharmacology
- Inhibition of neuronal reuptake of serotonin
- Potentiation of neurotransmitter's action
- Rapid absorption with maximum plasma concentrations occurring approximately 1-2 hours after tablet intake
- Absolute bioavailability is 42%
- Rapid distribution with a mean steady state volume of distribution of 162 L
- Extensive metabolism to multiple metabolites primarily through biotransformational pathways
Dosage & Administration
- Recommended starting dose for all patients is 30 mg
- Maximum recommended dosing frequency is once every 24 hours
- Dose may be increased to the maximum recommended dose of 60 mg if 30 mg is insufficient
- Not recommended for use in individuals below 18 years of age
- Safety and efficacy have not been established in patients age 65 years and over
- Caution is advised in patients with mild or moderate renal impairment
- Contraindicated in patients with moderate and severe hepatic impairment
Interaction
- Caution advised with CNS active medicinal products
- Prescribed with caution in patients who use PDE5 inhibitors
- Prescribed with caution in patients who use alpha adrenergic receptor antagonists
- Caution advised when used in patients taking Warfarin chronically
- Concomitant use of alcohol may increase chance or severity of adverse reactions
Contraindications
- Known hypersensitivity to Dapoxetine Hydrochloride
- Significant pathological cardiac conditions
- Concomitant treatment with monoamine oxidase inhibitors (MAOIs), thioridazine
- Concomitant treatment with serotonin reuptake inhibitors or other medicinal/herbal products with serotonergic effects
- Use with recreational drugs

Pregnancy & Lactation
- Not indicated for use by women
- Unknown excretion through human breast milk
Precautions & Warnings
- Patients should be advised not to use in combination with recreational drugs
- Combining alcohol with the drug may increase neurocognitive effects
- Patients need to be made aware of the possibility of experiencing syncope during treatment
- An orthostatic test should be performed before initiating therapy
- Caution is advised in patients taking medicinal products known to affect platelet function or anticoagulants
- Prescribed with caution in patients who use alpha adrenergic receptor antagonists
Overdose Effects
- No case of overdose reported
- No unexpected adverse events with daily doses up to 240 mg
- Symptoms of overdose with SSRIs include serotonin-mediated adverse reactions such as somnolence, gastrointestinal disturbances, tachycardia, tremor, agitation and dizziness
- No evidence of withdrawal syndrome
