Indications
- Treatment of premature ejaculation (PE) in men 18 to 64 years of age
- Persistent or recurrent ejaculation with minimal sexual stimulation
- Marked personal distress or interpersonal difficulty as a consequence of PE
- Poor control over ejaculation
Pharmacology
- Inhibition of neuronal reuptake of serotonin
- Potentiation of neurotransmitter's action at pre-and post synaptic receptors
- Mediated by the sympathetic nervous system
- Rapid absorption with maximum plasma concentrations
Dosage & Administration
- Recommended starting dose for all patients is 30 mg
- Maximum recommended dosing frequency is once every 24 hours
- Not recommended for use in individuals below 18 years of age
- Caution advised in patients with mild or moderate renal impairment
Interaction
- Concomitant use with CNS active medicinal products
- Prescribed with caution in patients who use PDE5 inhibitors
- Possible reduced orthostatic tolerance in patients using alpha adrenergic receptor antagonists
- Caution when used in patients taking Warfarin chronically
Contraindications
- Known hypersensitivity to Dapoxetine Hydrochloride
- Significant pathological cardiac conditions
- Concomitant treatment with monoamine oxidase inhibitors (MAOIs) or serotonin reuptake inhibitors
- Contraindicated during concomitant treatment with thioridazine

- Dry mouth
- Headache
- Diarrhea
- Insomnia
- Low rates of sexual dysfunction
- Very mild adverse effects on loss of libido and ED
Pregnancy & Lactation
- Not indicated for use by women
- Unknown excretion through human breast milk
Precautions & Warnings
- Patients advised not to use Susten in combination with recreational drugs
- Avoiding alcohol while taking Susten
- Possibly prodromal symptoms of syncope
- Oral orthostatic test before initiating therapy
- Caution in patients with a history of bleeding or coagulation disorders
Overdose Effects
- No reported case of overdose
- Symptoms of overdose with SSRIs could include serotonin-mediated adverse reactions
- No evidence of withdrawal syndrome in a clinical trial
