Brezofil 200 mg (Tablet)

Indications

• Bronchial asthma
• Bronchospasm
• Chronic obstructive pulmonary disease (COPD)
• Pulmonary disease with spastic bronchial component

Pharmacology

• Novel bronchodilator
• Selective inhibition of phosphodiesterase 4
• Relaxation of bronchial smooth muscle
• Inhibition of platelet activating factor (PAF) and leukotriene production

Dosage & Administration

• Elderly dosage: 200 mg tablet two or three times daily
• Adults dosage: 400 mg tablet two or three times daily
• Children dosage: 10 ml syrup or 200 mg tablet for >12 years of age
• Children dosage: 6-9 mg/kg body weight for 6-12 years of age
• Doxophylline SR tablet for required daily dose of 400 mg

Interaction

• Not to be administered with other xanthine preparations
• Limit consumption of caffeine-containing beverages and food
• Caution with ephedrine or sympathomimetic drugs
• Possible hepatic clearance reduction with certain drugs like erythromycin, TAO, lincomycin, etc.
• Phenytoin and other anticonvulsants may increase clearance

Contraindications

• Known hypersensitivity to the drug or other xanthine derivatives
• Acute myocardial infarction
• Hypotension
• Lactation

Side Effects

• Nausea
• Vomiting
• Epigastric pain
• Headache
• Irritability
• Insomnia
• Tachycardia
• Tachypnea
• Hyperglycemia
• Albuminuria
• Severe cardiac arrhythmias in case of overdose
• Tonic-clonic seizure in case of overdose

Pregnancy & Lactation

• Animal tests show no interference with pre and postnatal growth
• Use during pregnancy requires careful evaluation
• Contraindicated during lactation
• No effect on ability to drive or use machines

Precautions & Warnings

• Factors reducing hepatic clearance may increase plasma levels
• Factors include age, congestive cardiac decompensation, chronic obstructive pulmonary disease, severe liver disease, concomitant infections, and certain drug concurrent administration
• Recommended monitoring of blood levels in case of clearance-affecting factors
• Caution in patients with various conditions such as cardiac disease, hypertension, hypoxemia, hyperthyroidism, liver disease, renal impairment
• No risk of addiction or dependence

Overdose Effects

• Severe cardiac arrhythmias
• Tonic-clonic seizure
• Discontinuation of treatment may be necessary
• Symptomatic treatment of cardiovascular collapse in case of overdose
