Dosage & Administration
- Should be taken 15-30 minutes before meals
- Usual recommended oral dose for adults: 10-20 mg, every 6-8 hours daily
- Maximum adult dose: 80 mg daily
- Usual recommended dose for children: 2-4 ml suspension/10 kg body weight, every 6-8 hours daily

- Dyspeptic symptom complex
- Acute nausea and vomiting
- Parkinson's disease
- Radiological studies
Pharmacology
- Dopamine antagonist
- Blocks dopamine receptors in CTZ and stomach
- Restores normal motility of upper gastrointestinal tract
- Facilitates gastric emptying
- Enhances antral and duodenal peristalsis
Interaction
- Concomitant administration of anticholinergic drugs may antagonize the effect of domperidone
- Antacids and antisecretory drugs should not be given simultaneously with domperidone
- Main metabolic pathway is through CYP3A4 enzyme
- May interact with neuroleptics and dopaminergic agonists
Contraindications
- Known hypersensitivity to the drug
- Neonates
- Gastrointestinal hemorrhage, mechanical obstruction or perforation
- Prolactin releasing pituitary tumor
Side Effects
- Transient intestinal cramps
- Rare extrapyramidal phenomena
- Possible increase in plasma prolactin level
- Rare allergic reactions such as rash and urticaria
Pregnancy & Lactation
- Should only be used during the first trimester if justified by therapeutic benefit
- Excreted in breast milk but concentrations are 4 times lower than plasma
- Not recommended for nursing mothers unless benefits outweigh risks

- Use with caution in children due to risk of extrapyramidal reactions
- Use with caution in patients with hepatic impairment

- Use in infants with caution and under close medical supervision
- Use with caution in patients with liver or kidney disorders
Overdose Effects
- Symptoms may include drowsiness, disorientation, and extrapyramidal reactions
- Administration of activated charcoal and close observation recommended
