Indications
- Bronchial asthma
- Bronchospasm

Pharmacology
- Novel bronchodilator
- Selective inhibition of phosphodiesterase 4
- Relaxation of bronchial smooth muscle
- Inhibition of platelet activating factor (PAF)
- Inhibition of leukotriene production
Dosage & Administration
- 200 mg tablet for elderly two or three times daily
- 400 mg tablet for adults two or three times daily
- 10 ml syrup or 200 mg tablet for >12 years of age
- 6-9 mg/kg body weight for 6-12 years of age
- Doxophylline SR tablet for required 400 mg daily dose
Interaction
- Avoid administration with other xanthine preparations
- Limit consumption of caffeine-containing beverages and food
- Exercise caution with ephedrine or other sympathomimetic drugs
- Concurrent administration with erythromycin, TAO, lincomycin, clindamycin, allopurinol, cimetidine, influenza vaccine, and propanolol may reduce hepatic clearance
- Phenytoin, other anticonvulsants, and cigarette smoking may increase clearance
Contraindications
- Known hypersensitivity to doxophylline or other xanthine derivatives
- Acute myocardial infarction
- Hypotension
- During lactation

- Epigastric pain
- Headache
- Irritability
- Insomnia
- Tachycardia
- Extrasystoles
- Tachypnea
- Rare cases of hyperglycemia or albuminuria
- Severe cardiac arrhythmias in case of overdose
- Tonic-clonic seizure in case of overdose
Pregnancy & Lactation
- Animal tests show no interference with pre and postnatal growth
- Insufficient clinical evidence for use during pregnancy
- Contraindicated during lactation
- Effects on ability to drive and use machines: Does not affect alertness
Precautions & Warnings
- Factors that may reduce hepatic clearance
- Caution in patients with cardiac disease, hypertension, chronic obstructive pulmonary disease, severe liver disease, and concomitant infections
- Monitoring of blood levels recommended in certain cases
Overdose Effects
- Severe cardiac arrhythmias
- Tonic-clonic seizure
- Absence of specific antidote
