Indications

  • Bronchial asthma
  • Bronchospasm
  • Chronic obstructive pulmonary disease (COPD)
  • Pulmonary disease with spastic bronchial component

Pharmacology

  • Novel bronchodilator
  • Selective inhibition of phosphodiesterase 4
  • Relaxes bronchial smooth muscle
  • Decreased affinities toward adenosine A1 and A2 receptors
  • Inhibition of platelet activating factor (PAF) and leukotriene production

Dosage & Administration

  • Elderly dosage: 200 mg tablet two or three times daily
  • Adults dosage: 400 mg tablet two or three times daily
  • Children dosage: varies by age and body weight
  • Special dose for daily 400 mg requirement

Interaction

  • Contraindicated with other xanthine preparations
  • Limit consumption of caffeine-containing beverages and food
  • Caution with ephedrine or other sympathomimetic drugs
  • Concurrent administration with erythromycin, TAO, lincomycin, etc.
  • Increase dosage with phenytoin, anticonvulsants, and cigarette smoking

  • Hypersensitivity to the drug or other xanthine derivatives
  • Acute myocardial infarction
  • Hypotension
  • Lactation

Precautions & Warnings

  • Factors reducing hepatic clearance of xanthine derivatives
  • Caution with cardiac disease, hypertension, hypoxemia, hyperthyroidism
  • Monitoring blood levels in cases of influencing factors

Overdose Effects

  • Severe cardiac arrhythmias
  • Tonic-clonic seizure
  • Symptomatic treatment of cardiovascular collapse
