Dosage
- 10-20 mg (1-2 tablet or 10-20 ml suspension) every 6-8 hours daily for adults
- 2-4 ml suspension/10 kg body weight or 0.4-0.8 ml pediatric drops/10 kg body weight every 6-8 hours daily for children
- 30-60 mg every 4-8 hours for adults via rectum
- Maximum dose of 80 mg daily for adults
- Maximum daily rectal dose of 30 mg for children weighing 10-25 kg

- Acute nausea and vomiting
- Parkinson's disease
- Radiological studies
Pharmacology
- Dopamine antagonist
- Blocks dopamine receptors in the ChemoreceptorTrigger Zone (CTZ) and stomach
- Restores normal motility and tone of the upper gastrointestinal tract
- Facilitates gastric emptying
- Enhances antral and duodenal peristalsis
- Regulates contraction of the pylorus
- Increases esophageal peristalsis
Interaction
- Concomitant administration of anticholinergic drugs may antagonize the antidyspeptic effect of domperidone
- Antacids and antisecretory drugs should not be given simultaneously with domperidone
- Main metabolic pathway of domperidone is through CYP3A4
- In vitro data suggest that concomitant use of drugs that significantly inhibit CYP3A4 may increase plasma levels of domperidone
Contraindications
- Known hypersensitivity to domperidone
- Neonates
- Gastrointestinal hemorrhage
- Mechanical obstruction or perforation
- Prolactin releasing pituitary tumor (prolactinoma)
Side Effects
- Transient intestinal cramps
- Rare extrapyramidal phenomena
- Increase in plasma prolactin level
- Rare allergic reactions such as rash and urticaria
Pregnancy & Lactation
- Use during the first trimester of pregnancy if justified by therapeutic benefit
- No evidence of increase in the risk of malformations in humans
- Excreted in breast milk with concentrations 4 times lower than corresponding plasma concentrations
Precautions & Warnings
- Use with caution in children due to increased risk of extrapyramidal reactions
- Use with caution in patients with hepatic impairment
Use in Special Populations
- Use in infants with caution and under close medical supervision due to immature blood-brain barrier
- Use with caution in patients with hepatic impairment
- In patients with severe renal insufficiency, elimination half-life of domperidone is increased
- Dosing frequency should be reduced in patients with renal insufficiency
Overdose Effects
- Symptoms may include drowsiness, disorientation, and extrapyramidal reactions
- Activated charcoal administration and close patient observation recommended

Indications Details

Mode of Administration
