Rimactazid 450 mg+300 mg (Tablet)

Indications

• Continuation phase treatment of tuberculosis

Pharmacology

• Active bactericidal anti-TB drugs
• Active against rapidly growing extracellular organisms
• Bactericidal activity intracellularly
• Rifampicin inhibits DNA-dependent RNA polymerase activity
• Isoniazid acts against actively growing tubercle bacilli

Dosage & Administration

• For body weight <50 kg: 3 tablets of 150/100 once daily or 1 tablet of 450/300 once daily
• For body weight >50 kg: 2 tablets of 300/150 once daily
• Max dosage for hepatic impairment: 8 mg/kg daily

Interaction

• May reduce effectivity of hormonal contraceptives
• Reduced absorption with antacids
• May decrease plasma concentrations of various antivirals and anticonvulsants
• Increased risk of hepatotoxicity with halothane
• May inhibit the metabolism of certain anticonvulsants and benzodiazepines
• May enhance the CNS effects of certain medications
• Loss of glucose control in patients on oral hypoglycemics with isoniazid

Contraindications

• Hypersensitivity to rifamycins and/or to INH
• Acute liver diseases
• Peripheral neuritis

• Reddish discoloration of body fluids
• Hepatic effects
• Renal effects
• Gastrointestinal effects
• Central and peripheral nervous system effects
• Haematological changes
• Effects on skin and appendages
• Endocrine effects

Pregnancy & Lactation

• Pregnancy Category C
• Animal reproduction studies have shown adverse effects

Precautions & Warnings

• Careful monitoring in daily users of alcohol
• Careful monitoring in patients with active chronic liver disease or severe renal dysfunction
• Careful monitoring in individuals over age 35
• Careful monitoring in concurrent use of any chronically administered medication
• Careful monitoring in history of previous discontinuation of isoniazid
• Careful monitoring in existence of peripheral neuropathy or conditions predisposing to neuropathy
• Careful monitoring in pregnancy
• Careful monitoring in injection drug use
• Careful monitoring in women belonging to minority groups, particularly in the post-partum period
• Careful monitoring in HIV seropositive patients
