Indications
- Palliative treatment of advanced carcinoma of the breast
- Palliative treatment of advanced carcinoma of the endometrium
- Treatment of anorexia in patients with a diagnosis of Acquired Immunodeficiency Syndrome (AIDS) & cancer
- Treatment of cachexia in patients with a diagnosis of Acquired Immunodeficiency Syndrome (AIDS) & cancer
- Treatment of unexplained, significant weight loss in patients with a diagnosis of Acquired Immunodeficiency Syndrome (AIDS) & cancer
Pharmacology
- Synthetic, antineoplastic and progestational drug
- Mechanism of action against endometrial carcinoma is not fully known
- Inhibition of pituitary gonadotrophin production
- Decrease in estrogen secretion
- Antineoplastic action on breast carcinoma by modifying the action of other steroid hormones
- Direct cytotoxic effect on tumor cells
- Estrogen receptor mechanism
- Plasma half-life of 33 to 38 hours
Dosage & Administration
- Tablet dosage for breast cancer: 160 mg/day
- Tablet dosage for endometrial carcinoma: 40-320 mg/day in divided doses
- At least 2 months of continuous treatment is considered adequate for determining efficacy
- Oral suspension initial adult dosage: 800 mg/day (20 ml/day)
Interaction
- Possible interactions with concomitant medications not investigated
- Estrogens and progestogens may cause fluid retention
- Therapy with these agents should be administered cautiously in patients with preexisting problems with excess fluid
Contraindications
- History of hypersensitivity to Megestrol Acetate or any component of the formulation
- Known or suspected pregnancy

Pregnancy & Lactation
- Pregnancy Category D
- Use during the first four months of pregnancy not recommended
- Very small amounts excreted in mother's milk
- Harmful effect on the newborn not known
- Nursing should be discontinued during treatment with Megestrol Acetate
Precautions & Warnings
- Close surveillance for recurrent or metastatic cancer patients
- Use with caution in patients with a history of thromboembolic disease
- Exacerbation of preexisting diabetes with increased insulin requirements
Use in Special Populations
- Safety and effectiveness in pediatric patients not established
- Insufficient data from clinical studies available for patients 65 years of age and older
Overdose Effects
- No serious unexpected side effects reported from dosages as high as 1600 mg/day
