Indications
- Treatment of active duodenal ulcer
- Treatment of benign gastric ulcer
- Treatment and prevention of ulcer associated with non-steroidal anti-inflammatory agent

- Zollinger-Ellison Syndrome treatment
- Gastroesophageal reflux disease (GERD) treatment
- Gastrointestinal haemorrhage from stress ulcer in seriously ill patient treatment
- Recurrent haemorrhage in patients with bleeding peptic ulcer treatment
- Prevention of acid aspiration before general anesthesia
- Treatment of episodic dyspepsia
Mode of Action
Competitively blocks histamine at H2-receptors of the gastric parietal cells inhibiting gastric acid secretion
Pharmacology
Does not affect pepsin secretion, pentagastrin-stimulated intrinsic factor secretion or serum gastrin
Dosage and Administration
- Usual 150 mg dosage twice daily for 4 to 8 weeks for duodenal and gastric ulcer
- 150 mg twice daily or 300 mg at bedtime for reflux esophagitis
- 150 mg 3 times daily for Zollinger-Ellison Syndrome
- 150 mg twice daily for episodic dyspepsia
Interaction
- Delayed absorption and increased peak serum concentration with propantheline bromide
- Minimally inhibits hepatic metabolism of coumarin anticoagulants, theophylline, diazepam, and propranolol
- May alter absorption of pH-dependent drugs like ketoconazole, midazolam, and glipizide
- May reduce bioavailability with antacids
