Indications
- Reduction in Post-Prandial Hyperglycaemia
- Non-insulin-dependent diabetes mellitus (NIDDM) treatment
- Combination therapy with other Oral Hypoglycaemic Agents (OHAs)
- In elderly patients and those with hepatic dysfunction or renal impairments
- Prevention of onset of type 2 diabetes mellitus in impaired glucose tolerance
- Treatment of glycogen storage disease
- Treatment of non-diabetic Hyperinsulinemia
- Prevention of hypoglycaemic attacks in steroid induced diabetes mellitus
Description
- Selective inhibition of α-glucosidase in the enteric canal
- Delays digestion and absorption of carbohydrate
- Suppression of sharp increase in post-prandial plasma glucose
Pharmacology
- Competitive inhibitors of alpha-glucosidase enzymes
- Inhibition of membrane-bound intestinal alpha-glucosidases
- Inhibition of pancreatic alpha-amylase
- Reduction in the rate of digestion of complex carbohydrates
Dosage & Administration
- Orally administered in a single dose of 0.2 mg, 3 times a day
- Pediatrics: Safety and effectiveness not established
- Geriatrics: Desirable to start at a lower dose
Interaction
- Should be administered with care when co-administered with antidiabetic drugs
- Drugs enhancing the hypoglycaemic action
- Drugs diminishing the hypoglycaemic action
Contraindications
- Hypersensitivity to Voglibose or excipients
- Diabetic ketoacidosis or pre-coma
- Severe infection, operation, or serious trauma
- Gastrointestinal obstruction or predisposition to it
Side Effects
- Diarrhoea, loose stools, abdominal pain
- Constipation, loss of appetite, nausea, vomiting, heartburn
- Increased gas, intestinal obstruction-like symptoms
- Hypoglycaemia with OHAs plus Vibose
- Delay in digestion and absorption of disaccharides
- Fulminant hepatitis, serious liver dysfunction, jaundice
- Numbness, edema, blurred vision, malaise, weakness
- Hyperkalemia, increased pancreatic enzyme (serum amylase)
Pregnancy & Lactation
- Safety in pregnancy not established
- Recommended to avoid administration in lactating and nursing mothers
Precautions & Warnings
- Careful administration in patients receiving other antidiabetic drugs
- Caution in patients with a history of laparotomy or ileus
- Special attention in patients with chronic intestinal disease
- Close observation in patients with serious hepatic or renal dysfunction
- Elderly patients need careful administration
Use in Special Populations
- Poorly absorbed after oral doses with negligible renal excretion
Overdose Effects
- Transient increase in flatulence, diarrhoea, and abdominal discomfort
- No serious systemic reactions expected in the event of an overdose
