Indications

Pharmacology
- Proton pump inhibitor
- Suppression of gastric acid secretion
- Specific inhibition of the H+/K+-ATPase
- Peak plasma levels occur at approximately 1.5 hours after oral administration
- Extensively metabolized in the liver
- Plasma protein binding is constant over the concentration range
- The plasma elimination half-life is approximately 1–1.5 hours
Dosage
- 20 mg or 40 mg Once Daily for 4-8 Weeks for Healing of Erosive Esophagitis
- 20 mg Once Daily for Maintenance of Healing of Erosive Esophagitis
- 20 mg Once Daily for Symptomatic GERD
- Triple Therapy for Helicobacter pylori eradication
- 20-80 mg Once Daily for Zollinger-Ellison Syndrome
- 20-40 mg Once Daily for Acid-related Dyspepsia
- 20 mg Once Daily for 2-4 weeks for Duodenal ulcer
Administration
- Tablet or capsule should be swallowed whole and taken one hour before a meal
- Oral Suspension should be taken after mixing with water
- IV Injection: 20 mg or 40 mg for adult patients, dosages for pediatric patients
- IV Infusion for risk reduction of rebleeding of Gastric or Duodenal Ulcers following therapeutic endoscopy in adults
Interaction
- Extensively metabolized by CYP2C19 and CYP3A4
- No clinically significant interactions with certain drugs
- Potential interference with CYP2C19 and gastric pH-dependent drugs
- Increased plasma levels when co-administered with clarithromycin and amoxicillin
Contraindications
- Known hypersensitivity to any of the formulation

Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women
- No teratogenic effects in animal studies
- Excretion in milk not studied, breastfeeding should be discontinued if essential
Precautions & Warnings
- Symptomatic response does not preclude the presence of gastric malignancy
- Instructions for patients on capsule administration
- Use of antacids while taking Esotor
Use in Special Populations
- No established safety and effectiveness in pediatric patients
- No overall differences in safety and efficacy observed in the elderly
- No dosage adjustment recommended for patients with mild to moderate hepatic insufficiency
- Pharmacokinetics not expected to be altered in renal impairment
Overdose Effects
- Acute toxicity signs in animal studies
- No specific antidote known
- Treatment should be symptomatic and supportive
