Pharmacology

Dosage & Administration
- Adult dosage: 1 film-coated tablet twice daily
- Children 3-7 years dosage: 1 effervescent tablet daily
- Children 7 years and older dosage: 1 to 2 effervescent tablets daily
- Best taken with or just after main meals with a full glass of water
Interaction
- Consideration of hypercalcemia risk in patients taking thiazide diuretics
- Potential reduction of Calcium absorption by certain foods
- Interactions with phenytoin, barbiturates, glucocorticoids, and cardiac glycosides
- Medical supervision needed in certain cases
Contraindications
- Absolute contraindications: hypercalcaemia, primary hyperparathyroidism, Vitamin-D3 overdosage, severe renal failure
- Relative contraindications: osteoporosis due to prolonged immobilisation, renal stones, severe hypercalciuria
- Hypersensitivity to any tablet ingredients
Side Effects
- Mild gastro-intestinal disturbances
- Occasional skin rash with vitamin-D3 supplements
- Hypercalciuria and rare cases of hypercalcaemia with long-term treatment at high doses
Pregnancy & Lactation
- Increased requirements for calcium and vitamin-D3 during pregnancy and lactation
- Caution with vitamin-D3 overdoses during pregnancy
- Transfer of vitamin-D3 and its metabolites into breast milk
Precautions & Warnings
- Careful supervision in patients with mild to moderate renal failure or mild hypercalciuria
- Periodic checks of plasma calcium levels and urinary calcium excretion
- Monitoring of serum and urinary calcium levels and kidney function with long-term treatment
Overdose Effects
- Symptoms of overdosage include nausea, vomiting, drowsiness, dry mouth, loss of appetite, and more
