Anastrol 1 mg (Tablet)

Indications

• Adjuvant treatment of postmenopausal women with hormone receptor-positive early breast cancer
• First-line treatment for postmenopausal women with hormone receptor-positive or hormone receptor unknown locally advanced or metastatic breast cancer
• Second-line treatment of advanced breast cancer in postmenopausal women with disease progression following tamoxifen therapy

Pharmacology

• Non-steroidal aromatase inhibitor
• Inhibition of aromatase enzyme to suppress estrogen biosynthesis

Dosage & Administration

• One tablet 1 mg taken once a day
• Continued use for advanced breast cancer until tumor progression

Interactions

• Reduced plasma concentration when co-administered with tamoxifen
• Should not be administered with tamoxifen or estrogen-containing therapies
• No significant impact on other drugs through cytochrome P450 inhibition

Contraindications

• Contraindicated in pregnancy and premenopausal women
• Contraindicated in patients with hypersensitivity to the drug or its excipients

Side Effects

• Serious adverse reactions include skin reactions, allergic reactions, and liver function changes
• Common adverse reactions include hot flashes, asthenia, arthritis, nausea, rash, osteoporosis, fractures, back pain, insomnia, headache, and more

Pregnancy & Lactation

• Pregnancy Category X
• Not known if excreted in human milk, decision should be made on nursing

Precautions & Warnings

• Increased risk of ischemic cardiovascular events in patients with pre-existing ischemic heart disease
• Decrease in bone mineral density compared to baseline, higher incidence of elevated cholesterol compared to tamoxifen

Overdose Effects

• Well tolerated at doses up to 60 mg in healthy male volunteers
• No specific antidote, treatment must be symptomatic
