Indications
- Treatment of newly diagnosed glioblastoma multiforme
- Treatment of refractory anaplastic astrocytoma
Pharmacology
- Temozolomide undergoes rapid nonenzymatic conversion to MTIC
- Cytotoxicity of MTIC is due to alkylation of DNA
Dosage
- Initial dose for newly diagnosed GBM: 75 mg/m2 for 42 days concomitant with radiotherapy
- Initial dose for refractory anaplastic astrocytoma: 150 mg/m2 once daily for 5 consecutive days
Administration
- Take on an empty stomach
- Take at least 1 hr before meals
Interaction
- Administration of valproic acid decreases oral clearance of Temonix
Contraindications
- History of hypersensitivity reaction to any of its components
- History of hypersensitivity to DTIC

Precautions & Warnings
- Monitor Absolute Neutrophil Count (ANC) and platelet count
- Prophylaxis required for Pneumocystis carinii pneumonia (PCP)
- Fetal harm can occur when administered to a pregnant woman
Use in Special Populations
- Safety and effectiveness in pediatric patients have not been established
- Caution should be exercised when administered to patients with severe renal impairment
- Caution should be exercised when administered to patients with severe hepatic impairment
Overdose Effects
- Dose-limiting toxicity was hematologic
- Overdose of 2000 mg per day for 5 days reported adverse reactions
