Indications
- Newly Diagnosed Glioblastoma Multiforme
- Refractory Anaplastic Astrocytoma
Pharmacology
- Rapid nonenzymatic conversion at physiologic pH
- Cytotoxicity of MTIC
- Alkylation of DNA
Dosage
- Newly Diagnosed GBM: 75 mg/m^2 for 42 days concomitant with focal radiotherapy
- Refractory Anaplastic Astrocytoma: Initial dose 150 mg/m^2 once daily for 5 consecutive days per 28-day treatment cycle
Administration
- Should be taken on an empty stomach
- Take at least 1 hr before meals
Interaction
- Valproic Acid: Administration decreases oral clearance of Temonix by about 5%
Contraindications
- History of hypersensitivity reaction to any of its components
- History of hypersensitivity to DTIC

Precautions & Warnings
- Myelosuppression: monitor Absolute Neutrophil Count (ANC) and platelet count
- Pneumocystis carinii pneumonia (PCP) prophylaxis required
- Fetal harm can occur when administered to a pregnant woman
Use in Special Populations
- Pediatric Use: Safety and effectiveness not established
- Caution should be exercised when administered to patients with severe renal impairment
- Caution should be exercised when administered to patients with severe hepatic impairment
Overdose Effects
- Dose-limiting toxicity was hematologic
- Adverse reactions after overdose include bone marrow suppression and multi-organ failure
