Pharmacology

Cisplatin produces intra-and interstrand cross links modifying DNA structure and inhibits DNA synthesis. It also inhibits protein and RNA synthesis.

Dosage & Administration

Administered by slow intravenous infusion
Dosage varies based on the type of cancer being treated

Interaction

Additive toxicity with antineoplastic drugs
Interactions with nephrotoxic, ototoxic, renally excreted, anticonvulsant, and antigout drugs

Contraindications

Pre-existing renal impairment
Hypersensitivity to Cisplatin or Platinum containing compounds
Myelosuppresed patients
Patients with hearing impairment

Side Effects

Severe nausea and vomiting
Electrolyte disturbances (hypomagnesemia, hypocalcemia, hypokalemia)
Renal tubular dysfunction
