Indications

Pharmacology

Cisplatin produces intra-and interstrand cross links modifying DNA structure and inhibits DNA synthesis. It also inhibits protein and RNA synthesis.

Dosage & Administration

  • Administered by slow intravenous infusion
  • Dosage varies based on the type of cancer being treated

Interaction

  • Additive toxicity with antineoplastic drugs
  • Interactions with nephrotoxic, ototoxic, renally excreted, anticonvulsant, and antigout drugs

Contraindications

  • Pre-existing renal impairment
  • Hypersensitivity to Cisplatin or Platinum containing compounds
  • Myelosuppresed patients
  • Patients with hearing impairment

Side Effects

  • Severe nausea and vomiting
  • Electrolyte disturbances (hypomagnesemia, hypocalcemia, hypokalemia)
  • Renal tubular dysfunction
