Pharmacology
Cisplatin produces intra-and interstrand cross links modifying DNA structure and inhibits DNA synthesis. It also inhibits protein and RNA synthesis.
Dosage & Administration
- Administered by slow intravenous infusion
- Dosage varies based on the type of cancer being treated
Interaction
- Additive toxicity with antineoplastic drugs
- Interactions with nephrotoxic, ototoxic, renally excreted, anticonvulsant, and antigout drugs
Contraindications
- Pre-existing renal impairment
- Hypersensitivity to Cisplatin or Platinum containing compounds
- Myelosuppresed patients
- Patients with hearing impairment
Side Effects
- Severe nausea and vomiting
- Electrolyte disturbances (hypomagnesemia, hypocalcemia, hypokalemia)
- Renal tubular dysfunction
