Indications
- Carcinoma of the colon or rectum

Pharmacology
Fluorouracil is converted into the active 5-fluorodeoxyuridine monophosphate (FdUMP) by a variety of different metabolic pathways. The drug works by inhibiting the enzyme thymidylate kinase which results in reduced formation of thymidine and thus of DNA.
Dosage & Administration
- Intravenous delivery by rapid bolus injection or slow infusion
- Palliative management
- Poor risk patients and those who are not in an adequate nutritional state
- Maintenance therapy
- Usual adult dose for cervical cancer
- Usual pediatric dose for malignant disease
- Intra-arterial infusion
- Combination with radiation
- Combination with other anticancer drugs
Interaction
Pre-treatment with cimetidine for 4 weeks led to increased plasma concentrations of 5 FU PhaRes following intravenous and oral administration in 6 patients.
Contraindications
- Severely debilitated patients
- Patients with bone marrow suppression due to radiotherapy or chemotherapy
- Pregnant or breastfeeding women
- Patients without malignant illnesses

Precautions & Warnings
5-5 FU PhaRes is a highly toxic drug with a narrow margin of safety. Patients should be carefully supervised, and treatment should be discontinued promptly when signs of toxicity appear.

Overdose Effects
Cases of deliberate overdose are unknown but excessive toxicity because of the hematological effects. Supportive care is the main treatment for overdose.
Reconstitution
5-5 FU PhaRes is a cytotoxic anticancer drug and should be handled with caution. Special, trained personnel should reconstitute the drug.
Storage Conditions
Store the vial in original carton not exceeding 25℃. Do not refrigerate. Protect from light.
