Indications
- Adjuvant treatment of postmenopausal women with hormone receptor-positive early breast cancer
- First-line treatment of postmenopausal women with hormone receptor-positive or hormone receptor unknown locally advanced or metastatic breast cancer
- Second-line treatment of advanced breast cancer in postmenopausal women with disease progression following tamoxifen therapy
Pharmacology
Non-steroidal aromatase inhibitor
Dosage & Administration
One tablet 1 mg once a day
Hepatic Impairment
No changes in dose recommended for patients with mild-to-moderate hepatic impairment
Renal Impairment
No changes in dose necessary for patients with renal impairment
Pediatric Use
Safety and efficacy in pediatric patients not established
Geriatric Use
Pharmacokinetics not affected by age
Interaction
- Co-administration with tamoxifen reduces Aromatin plasma concentration by 27%
- Estrogen-containing therapies should not be used with Aromatin
Contraindications
- Pregnancy and Premenopausal Women
- Hypersensitivity to the drug or any excipients
Side Effects
- Serious adverse reactions include skin reactions, allergic reactions, changes in blood tests of the liver function
- Common adverse reactions include hot flashes, asthenia, arthritis, pain, hypertension, depression, nausea and vomiting, rash, osteoporosis, fractures, back pain, insomnia, headache, bone pain, peripheral edema, increased cough, dyspnea, pharyngitis, lymphedema
Pregnancy & Lactation
Pregnancy Category X, unknown if excreted in human milk
Precautions & Warnings
- Ischemic Cardiovascular Events observed in women with pre-existing ischemic heart disease
- Bone Effects include decrease in bone mineral density compared to baseline
- Elevated serum cholesterol reported during the ATAC trial

No specific antidote for overdosage, treatment must be symptomatic
