Indications
- Bronchial asthma
- Bronchospasm
- Chronic obstructive pulmonary disease (COPD)
- Pulmonary disease with spastic bronchial component
Pharmacology
- Novel bronchodilator
- Inhibits phosphodiesterase 4
- Relaxes bronchial smooth muscle
- Decreased affinities toward adenosine A1 and A2 receptors
Dosage & Administration
- Elderly: 200 mg tablet two or three times daily
- Adults: 400 mg tablet two or three times daily
- Children:
- >12 years of age: 10 ml syrup or 200 mg tablet two or three times daily
- 6-12 years of age: 6-9 mg/kg body weight two times daily
- Required daily dose of Doxophylline 400 mg: Doxophylline SR tablet to be taken once daily
Interaction
- Contraindicated with other xanthine preparations
- Limit consumption of beverages and food containing caffeine
- Exercise caution with ephedrine or other sympathomimetic drugs
- Concurrent administration with erythromycin, TAO, lincomycin, clindamycin, allopurinol, cimetidine, influenza vaccine, and propanolol may reduce hepatic clearance of xanthine derivatives

- Known hypersensitivity to the drug or other xanthine derivatives
- Acute myocardial infarction
- Hypotension
- Lactation

Pregnancy & Lactation
- Animal tests show no interference with pre and postnatal growth
- Use during pregnancy should be evaluated on a risk-benefit basis
- Contraindicated during lactation
- Does not affect the patient's alertness
Precautions & Warnings
- Factors may reduce hepatic clearance of xanthine derivatives
- Caution in patients with cardiac disease, hypertension, elderly, severe hypoxemia, hyperthyroidism, chronic corpulmonale, congestive heart failure, liver disease, peptic ulcer, and renal impairment
- Monitoring of the blood levels of the drug is recommended
- Caution in patients with congestive heart failure
- No risk of addiction or dependence
Overdose Effects
- Severe cardiac arrhythmias
- Tonic-clonic seizure
- Symptomatic treatment of cardiovascular collapse
