Indications
- Treatment of hypertension alone or with other antihypertensive agents
- Lowering blood pressure
- Initial therapy for patients likely to need multiple antihypertensive agents
Pharmacology
- Amlodipine as a dihydropyridine calcium channel blocker
- Amlodipine's effect on vascular smooth muscle and cardiac muscle cells
- Olmesartan Medoxomil's mechanism of blocking the vasoconstrictor and aldosterone-secreting effects of angiotensin II
Dosage & Administration
- Initial therapy with 5/20 mg once daily
- Titration up to a maximum of 10/40 mg once daily
- Adjusted dose for elderly patients
- Not recommended for patients >75 years old
Interaction
- Potential attenuation by NSAIDs including selective COX-2 inhibitors
- Monitoring of blood pressure, renal function, and electrolytes in patients on combination therapy
Contraindications
- Co-administration with Aliskiren in patients with diabetes

Precautions & Warnings
- Caution for hypotension in volume- or salt-depleted patients
- Risk of vasodilation in patients with severe aortic stenosis
- Possibility of increased frequency, duration, or severity of angina or acute MI in patients with severe obstructive coronary artery disease
Use in Special Populations
- Safety and effectiveness not established in pediatric patients
- No overall differences observed in subjects 65 years of age or older
- No studies in patients with renal impairment
- Initial therapy not recommended in hepatically impaired patients
