Composition

  • 375 mg tablet: Amoxicillin 250 mg, Clavulanic Acid 125 mg
  • 625 mg tablet: Amoxicillin 500 mg, Clavulanic Acid 125 mg
  • 1 gm tablet: Amoxicillin 875 mg, Clavulanic Acid 125 mg
  • Powder for Suspension: Amoxicillin 125 mg, Clavulanic Acid 31.25 mg
  • Powder for Suspension (Forte): Amoxicillin 400 mg, Clavulanic Acid 57.5 mg
  • 1.2 gm Injection: Amoxicillin 1 gm, Clavulanate Potassium 200 mg
  • 0.6 gm injection: Amoxicillin 500 mg, Clavulanate Potassium 100 mg

Pharmacology

  • Broad spectrum of bactericidal activity
  • Effective against Gram-positive & Gram-negative microorganisms
  • Inactivation of beta-lactamase enzymes
  • Optimized absorption at the start of a meal
  • Low levels of serum binding

Dosage

  • Varied dosages for different age groups
  • Different dosages for different types and severity of infections
  • Oral and intravenous administration
  • Duration of treatment not to exceed 14 days without review

Administration

  • Enhanced absorption when taken at the start of a meal
  • Intravenous injection over 2 minutes
  • Slow intravenous infusion over 30 minutes
  • Reconstituted vial to be used within 20 minutes

Interaction

  • Prolongation of bleeding time and prothrombin time
  • Reduction in efficacy of oral contraceptives
  • Potential interaction with allopurinol

Contraindications

  • History of Penicillin hypersensitivity
  • Possible cross-sensitivity with other beta-lactam antibiotics

Side Effects

  • Mild and transitory nature for most side effects
  • Possible gastrointestinal side effects
  • Rare reports of hepatitis and cholestatic jaundice
  • Rare occurrence of skin reactions

Pregnancy & Lactation

  • No teratogenic effect observed in animal studies
  • Limited use in human pregnancy with no untoward effect
  • Trace quantities detected in breast milk during lactation

Precautions & Warnings

  • Use with care in patients on anticoagulation therapy
  • Dosage adjustment for patients with renal or hepatic impairment
  • Maintenance of adequate fluid intake and urinary output

Use in Special Populations

  • Dosage adjustment for patients with renal impairment
  • Monitor hepatic function at regular intervals

Overdose Effects

  • Unlikely occurrence of overdose
  • Gastrointestinal symptoms and fluid/electrolyte imbalances may occur

Reconstitution

  • Reconstitution of IV injection with water for injection
  • Avoid mixing with specific fluids
  • Injection into infusion fluids containing glucose, bicarbonate, and dextran over 3-4 minutes

  • Reconstituted suspension to be refrigerated and used within 7 days
  • Reconstituted vial to be used within 20 minutes
