Indications

  • Prophylaxis of gastric and duodenal ulceration in NSAID users at high risk of complications
  • Healing of established NSAID-induced gastric and duodenal damage
  • Healing of gastric and duodenal ulcers in the absence of NSAID therapy
  • Induction of labor
  • Prevention and treatment of postpartum hemorrhage

Pharmacology

  • Extensively absorbed
  • Rapid de-esterification
  • Maximum plasma concentrations diminished when taken with food
  • Both antisecretory and mucosal protective properties
  • Increase in bicarbonate and mucus production

Dosage & Administration

  • 800 mcg daily for benign gastric and duodenal ulceration and NSAID-associated ulceration
  • 200 mcg 2-4 times daily for prophylaxis of NSAID-induced ulcer
  • 25 mcg vaginally for labor induction
  • 600 mcg orally for prevention of postpartum hemorrhage
  • 600-1000 mcg orally or 1000 mcg rectally for treatment of postpartum hemorrhage

Interaction

  • No clinically significant interaction with cardiac, pulmonary, CNS drugs, and NSAIDs
  • Decreased bioavailability with high doses of antacid

  • History of allergy to prostaglandins
  • Pregnancy
