Indications
- spasticity resulting from multiple sclerosis
- flexor spasms and concomitant pain, clonus and muscular rigidity
- skeletal muscle spasm resulting from rheumatic disorders
- spinal cord injuries and other spinal cord diseases
- cerebrovascular accidents or neoplastic or degenerative brain disease
Pharmacology
- inhibits monosynaptic and polysynaptic reflexes
- stimulates GABA receptors
- inhibits glutamate and aspartate release
- produces CNS depression
- exerts antinociceptive effect
Dosage & Administration
- 5 mg 3 times daily for adults & children over 10 years
- gradually increased dosage
- maximum daily dosage of 100 mg
- treatment initiation at 2.5 mg for children under 10 years
- gradual increase in dosage for children based on age
Interaction
- increased sedation with CNS agents, alcohol, or synthetic opiates
- interaction with antihypertensives leading to fall in blood pressure
- potentiation of effects with tricyclic antidepressants
- reports of mental confusion, hallucinations, and agitation with Parkinson's medication
- increased CNS depressant effects with MAO inhibitors
- potential synergistic effect with magnesium sulfate

Precautions & Warnings
- lower doses for impaired renal function or chronic hemodialysis
- cautious treatment for patients with psychotic disorders, schizophrenia, depressive or manic disorders
- use with caution in patients with epilepsy, peptic ulcers, cerebrovascular diseases, or hepatic, renal or respiratory failure
- respiratory and cardiovascular function monitoring
- potential improvement in neurogenic disturbances affecting bladder emptying
- poor tolerance and no significant benefit in stroke patients
- periodic laboratory tests for hepatic diseases or diabetes mellitus
Overdose Effects
- Gastric lavage in case of severe overdose
