Indications

  • Reduction of urate/uric acid formation
  • Management of gouty arthritis
  • Treatment of 2,8-dihydroxyadenine (2,8-DHA) renal stones
  • Management of recurrent mixed calcium oxalate renal stones

Pharmacology

  • Xanthine oxidase inhibitor
  • Orally administered
  • Reduces production of uric acid
  • Approximately 90% absorbed from the GI tract
  • Plasma half-life of about 1 to 2 hours

Dosage & Administration

  • Adults:
    • Low dosage introduction
    • Recommended dosage schedules based on condition severity
  • Children: Recommended dosage based on body weight
  • Elderly: Usage based on satisfactory urate reduction
  • Renal Impairment:
    • Reduced dosage for severe renal insufficiency
    • Use at longer intervals for severe renal impairment

Interaction

  • 6-Mercaptopurine or Azathioprine: Adjusted dosage when used concurrently
  • Vidarabine: Increased plasma half-life
  • Theophylline: Monitoring required when starting or increasing therapy
  • Ampicillin or Amoxicillin: Increase in frequency of skin rash reported
  • Ciclosporin: Possible increase in plasma concentration during concomitant treatment

  • Hypersensitivity to allopurinol
