Indications
- Reduction of urate/uric acid formation
- Management of gouty arthritis
- Treatment of 2,8-dihydroxyadenine (2,8-DHA) renal stones
- Management of recurrent mixed calcium oxalate renal stones
Pharmacology
- Xanthine oxidase inhibitor
- Orally administered
- Reduces production of uric acid
- Approximately 90% absorbed from the GI tract
- Plasma half-life of about 1 to 2 hours
Dosage & Administration
- Adults:
- Low dosage introduction
- Recommended dosage schedules based on condition severity
- Children: Recommended dosage based on body weight
- Elderly: Usage based on satisfactory urate reduction
- Renal Impairment:
- Reduced dosage for severe renal insufficiency
- Use at longer intervals for severe renal impairment
Interaction
- 6-Mercaptopurine or Azathioprine: Adjusted dosage when used concurrently
- Vidarabine: Increased plasma half-life
- Theophylline: Monitoring required when starting or increasing therapy
- Ampicillin or Amoxicillin: Increase in frequency of skin rash reported
- Ciclosporin: Possible increase in plasma concentration during concomitant treatment
