Indications

  • Gastric and duodenal ulcer
  • NSAID-associated duodenal and gastric ulcer
  • Prophylaxis in patients with a history of NSAID-associated duodenal and gastric ulcer
  • Gastro-esophageal reflux disease
  • Long-term management of acid reflux disease
  • Acid-related dyspepsia

Pharmacology

  • Inhibitor of gastric acid secretion
  • Blocker of hydrogen-potassium-adenosine triphosphatase (H+/K+ ATPase) enzyme system
  • Onset of antisecretory effect within one hour
  • Maximum effect occurring within two hours
  • Inhibition of secretion lasting up to 72 hours
  • Gradual return of secretory activity over 3 to 5 days after discontinuation

Contraindications

Side effects

  • Adverse reactions including skin rash, urticaria, pruritus, photosensitivity, bullous eruption, erythema multiforme, angioedema, alopecia, diarrhea, headache, constipation, nausea, vomiting, flatulence, abdominal pain
  • Rare symptoms such as paraesthesia, dizziness, light headedness, somnolence, insomnia, vertigo, reversible mental confusion, agitation, depression, hallucinations, arthritic and myalgic symptoms

Pregnancy and lactation

  • No adverse effects indicated on pregnancy or fetal health based on epidemiological studies
  • Suitability for use during pregnancy
  • Lack of information on passage into breast milk or effects on the neonate
  • Recommendation to discontinue breastfeeding if essential to use Omeprazole

Precautions and warnings

  • Avoid concomitant use with clopidogrel
  • Possible association with increased risk of osteoporosis-related fractures
  • Noted occurrence of atrophic gastritis in long-term treatment
  • Potential for methotrexate toxicities with concomitant use
