Indications
- Treatment of gastric and duodenal ulcer
- Treatment of NSAID-associated duodenal and gastric ulcer
- Prophylaxis in patients with history of NSAID-associated duodenal and gastric ulcer
- Treatment of gastro-esophageal reflux disease
- Long-term management of acid reflux disease
- Treatment of acid-related dyspepsia
- Treatment of severe ulcerating reflux esophagitis
- Prophylaxis of acid aspiration during general anesthesia
- Treatment of Zollinger-Ellison syndrome
- Treatment of Helicobacter pylori-induced peptic ulcer
Pharmacology
- Inhibitor of gastric acid secretion
- Blocks hydrogen-potassium-adenosine triphosphatase (H+/K+ ATPase) enzyme system in gastric parietal cell
- Onset of antisecretory effect occurs within one hour
- Maximum effect occurs within two hours
- Inhibition of secretion lasts up to 72 hours
- Return of secretory activity after discontinuation occurs over 3 to 5 days
Dosage
- Dosage varies based on the condition being treated
- Oral and IV administration
- Different doses for different conditions such as ulcers, reflux disease, and Zollinger-Ellison syndrome

Administration
- IV injection administration

Interaction
- Decreased intragastric acidity affecting ketoconazole absorption

Contraindications
- Contraindicated in patients with known hypersensitivity to omeprazole

Side Effects
- Mild and reversible adverse reactions
- Skin rash, urticaria, and pruritus
- Photosensitivity, bullous eruption, and erythema multiforme in isolated cases
- Diarrhea, headache, constipation, nausea/vomiting, flatulence, and abdominal pain
- Dizziness, light headedness, and feeling faint
- Reversible mental confusion, agitation, depression, and hallucinations
- Arthritic and myalgic symptoms
Pregnancy & Lactation
- No adverse effects on pregnancy or health of fetus/newborn child reported from studies
- No information available on passage into breast milk or effects on neonate
Precautions & Warnings
- Avoid concomitant use with clopidogrel
- Possibly associated with increased risk for osteoporosis-related fractures
- Atrophic gastritis noted occasionally in long-term treatment
- Concomitant use with methotrexate may lead to methotrexate toxicities
