Indications
- Reduction of urate/uric acid formation in conditions like gouty arthritis
- Management of 2,8-dihydroxyadenine renal stones
- Management of recurrent mixed calcium oxalate renal stones
Pharmacology
- Allopurinol is a xanthine oxidase inhibitor
- Reduces production of uric acid by inhibiting biochemical reactions
- Approximately 90% absorbed from the GI tract
Dosage & Administration
- Initial dosage of 100 mg/day for adults
- Dosage ranges for mild, moderately severe, and severe conditions
- Specific dosage for children and elderly
- Dosage adjustments for renal impairment
Interaction
- Reduced dose of 6-mercaptopurine or azathioprine when given concurrently with Duric
- Enhanced toxic effects with concomitant use of Duric and vidarabine
- Monitoring of theophylline levels during Duric therapy
- Increased frequency of skin rash with concurrent use of ampicillin or amoxicillin
- Possible increase in ciclosporin plasma concentration during concomitant treatment with Duric
Contraindications
- Contra-indicated in patients with known hypersensitivity to allopurinol

Overdose Effects
- Reported ingestion of up to 22.5 g without adverse effect
- Symptoms including nausea, vomiting, diarrhea, and dizziness
- Hydration to facilitate excretion of Duric and its metabolites
- Use of haemodialysis if necessary
