Indications
- Treatment of gastric and duodenal ulcer

Pharmacology
- Inhibitor of gastric acid secretion
- Blocks hydrogen-potassium-adenosine triphosphatase (H+/K+ ATPase) enzyme system
- Onset of antisecretory effect within one hour
- Maximum effect within two hours
- Inhibition of secretion lasts up to 72 hours
- Gradual return of secretory activity over 3 to 5 days upon discontinuation

- Oral administration for different conditions and durations
- IV Injection for prophylaxis of acid aspiration
- IV Injection for duodenal ulcer, gastric ulcer, or reflux oesophagitis
- IV Injection for Zollinger-Ellison syndrome

Administration
- Intravenous administration only
- Slow intravenous injection for IV Injection
- Intravenous infusion over a period of 20-30 minutes or more for IV Infusion
- Use only freshly prepared solution
- Solution to be used within specific timeframes
Interaction
- Potential interaction with ketoconazole
- Metabolized in the liver through cytochrome P450
- Potential interaction with diazepam, phenytoin, and warfarin
- Monitoring recommended for patients receiving warfarin or phenytoin
- Increased plasma concentrations during concomitant administration
- No evidence of interaction with certain drugs
Contraindications
- Contraindicated in patients with known hypersensitivity
- Possibility of malignancy should be excluded before treatment
Side Effects
- Mild and reversible adverse reactions
- Reported skin rash, urticaria, and pruritus
- Photosensitivity, bullous eruption, erythema multiforme, angioedema, alopecia reported in isolated cases
- Diarrhea, headache, constipation, nausea/vomiting, flatulence, abdominal pain reported
- Dry mouth, stomatitis, candidiasis reported as isolated cases
- Paraesthesia, dizziness, light headedness, feeling faint, somnolence, insomnia reported
- Reversible mental confusion, agitation, depression, hallucinations reported
- Arthritic and myalgic symptoms reported
Pregnancy & Lactation
- No adverse effects indicated
- Omeprazole can be used during pregnancy
- No information available on passage into breast milk or effects on neonate
Precautions & Warnings
- Avoid concomitant use of clopidogrel
- Increased risk for osteoporosis-related fractures
- Atrophic gastritis noted occasionally in long-term treatment
- Potential methotrexate toxicities with concomitant use of PPIs
