Pharmacology
- Potent H1 receptor antagonist
- No significant anticholinergic and antiserotonic effects
- No drowsiness effect
- Inhibits histamine-mediated early phase of allergic reaction
- Reduces migration of inflammatory cells
- Reduces release of mediators associated with late phase of allergic reaction
- Peak plasma concentration of 257 mcg/L within one hour
- Plasma half-life approximately 11 hours
- Consistent absorption from one subject to the next
- Renal clearance of 30 ml/minute
- Renal excretion half-life approximately 9 hours
Dosage and Administration
- Daily dosage for adults and children 6 years and older: 1 tablet or 2 teaspoonfuls
- Daily dosage for children 2-6 years: 1 teaspoonful once daily or 1/2 teaspoonful twice daily
- Daily dosage for children 6 months to 2 years: 1/2 teaspoonful once daily, with possibility of increased dose for children 12-23 months
- Single use injectable product for intravenous administration only
- Recommended dosages for adults, adolescents, and children 6 months to 11 years
- Not recommended for pediatric patients less than 6 years of age with impaired renal or hepatic function
Interaction
- No clinically significant drug interactions with Theophylline
- No clinically significant drug interactions with Azithromycin
- No clinically significant drug interactions with Pseudoephedrine
- No clinically significant drug interactions with Ketoconazole
- No clinically significant drug interactions with Erythromycin
- No significant drug interactions with other drugs
Contraindications
- Contraindicated in patients with history of hypersensitivity to Cetirizine or hydroxyzine
Side Effects
- Most common side effect: somnolence
Pregnancy and Lactation
- US FDA Pregnancy Category B
- No adequate and well-controlled studies in pregnant women
- Drug excreted in human milk
- Caution in administering to nursing women
Precautions and Warnings
- Caution required when driving a car or operating heavy machinery
Use in Special Populations
- Reduction in total body clearance and increase in AUC and half-life in patients with mild to moderate hepatic and renal impairment
- Safety demonstrated in pediatric patients aged 6 months to 5 years
- Reduction in clearance in elderly patients, proportionate to decrease in creatinine clearance
