Indications
- Schizophrenia and other psychoses
- Mania and hypomania
- Anxiety psychomotor agitation excitement
- Violent or dangerously impulsive behavior
- Intractable hiccup
- Nausea and vomiting of terminal illness
- Childhood schizophrenia
- Autism
Pharmacology
- Neuroleptic action
- Blockage of dopamine receptor
- Inhibition of hypothalamic and hypophyseal hormones
- Antiemetic properties
- Serotonin-blocking properties
- Weak antihistaminic properties
- Slight ganglion-blocking activity
Dosage
- Oral administration
- Intramuscular administration
- Initial adult dose: 25 mg three times daily or 75 mg at bedtime
- Dose increase by daily amounts of 25 mg
- Maximum daily dosage up to 1 g
- Dosage adjustment for children and elderly
Administration
May be taken with or without food. May be taken with meals to reduce GI discomfort.
Interaction
- Food, alcohol, and benztropine can reduce absorption
- Antacids can slow absorption
- Lithium and chronic administration of barbiturates can increase clearance
- Tricyclic antidepressants can decrease clearance
- Interactions with CYP1A2 inhibitors
Contraindications
- Circulatory collapse
- CNS depression
- Hypersensitivity to phenothiazines
- Bone marrow depression
- Phaeochromocytoma
- Hepatic failure or active hepatic disease
- Allergic-type reactions to excipients
