Indications

Pharmacology
- Active form: Calcitriol
- Binding to Vitamin D receptors (VDRs)
- Fat soluble with a half-life of 50 days
- Absorbed in the small intestine
- Metabolized to 25-hydroxy Vitamin D3 (Calcidiol)
- Second hydroxylation in the kidney to 1,25-dihydroxy Vitamin D3 (Calcitriol)
- Storage in tissues such as fat and muscle
- Elimination via faeces and urine
Dosage & Administration
- Dosage for adults: 40000 IU once weekly for 7 weeks for Vitamin D3 deficiency
- Maintenance therapy dose: 1400-2000 IU/day
- Dosage for children (12-18 years): 20000 IU once every 2 weeks for 6 weeks for Vitamin D3 deficiency
- Dosage for film-coated tablet: 1000 IU (1-2 tablets) daily
- Dosage for oroflash or chewable tablets: 1000-2000 IU daily
- Dosage for syrup for different age groups
- Dosage for injection for prevention and deficiency

Contraindications
- Contraindicated in diseases associated with hypercalcemia
- Contraindicated in known hypersensitivity to Vitamin D3
- Contraindicated in evidence of Vitamin D3 toxicity
Side Effects
- Symptoms such as anorexia, lassitude, nausea & vomiting, diarrhea, constipation, weight loss, polyuria, sweating, headache, thirst, vertigo
- Raised concentrations of calcium and phosphate in plasma and urine
Pregnancy & Lactation
- Safe use during pregnancy up to 4000 IU
- Recommended daily intake for pregnant women: 400 IU
- Excretion in breast milk
- Consideration of dose for breast-fed children

- Monitoring plasma-calcium concentration
