Indications
- Treatment of panic disorder with or without agoraphobia
- Adjunct in the treatment of Lennox-Gastaut Syndrome, akinetic and myoclonic seizures
- Patients with absence seizures who have failed to respond to succinimides
Pharmacology
- Exhibits anticonvulsive, sedative, muscle relaxing and anxiolytic effects
- Enhances GABAergic neurotransmission at inhibitory synapses
- Suppresses various types of paroxysmal activity including spike and wave discharge in absences seizures
DosageAndAdministration
- Initial dose for adults with seizure disorders should not exceed 1.5 mg/day
- Initial dose for adults with panic disorder is 0.25 mg given in two divided doses
- Dosage may be adjusted based on patient response
- Maximum recommended daily dose is 20 mg
Interaction
- Does not alter the pharmacokinetics of phenytoin, carbamazepine, or phenobarbital
Contraindications
- History of hypersensitivity to benzodiazepines
- Clinical or biochemical evidence of significant liver disease
- Acute narrow angle glaucoma

- Special attention required in infants and small children due to increased production of saliva and bronchial secretion
- Benzodiazepine pharmacologic effects appear to be greater in elderly patients
- No dose adjustment required in patients with renal impairment
OverdoseEffects
- Symptoms of overdose include drowsiness, ataxia, dysarthria, nystagmus
- May lead to coma, hypotension, cardiorespiratory depression
- Benzodiazepines increase the effects of other central nervous system depressants, including alcohol
