Indication
- Treatment of panic disorder with or without agoraphobia
- Treatment of Lennox-Gastaut Syndrome
- Treatment of akinetic and myoclonic seizures

Pharmacology
- Exhibits anticonvulsive, sedative, muscle relaxing and anxiolytic effects
- Enhancement of GABAergic neurotransmission at inhibitory synapses
- Effect on serotonin
Dosage and administration
- Adult initial dose not to exceed 1.5 mg/day divided into three doses
- Maintenance dosage individualized based on response
- Pediatric initial dose based on body weight and age
- Infant and children dosage for injection
- Adult dosage for injection and caution for rate of injection
Interaction
- No alteration of phenytoin, carbamazepine, or phenobarbital pharmacokinetics
- Effect on metabolism of other drugs not investigated
Contraindications
- History of hypersensitivity to benzodiazepines
- Clinical or biochemical evidence of significant liver disease
- Contraindicated in acute narrow angle glaucoma
Side effects
- CNS depression such as drowsiness and ataxia
- Behavior problems
- Abnormal eye movements, aphonia, coma, tremor, vertigo, confusion, depression, amnesia
Pregnancy and lactation
- Potential to produce congenital malformations
- Use only if benefits outweigh risks
- Breastfeeding not recommended
Precautions and warnings
- May increase incidence of generalized tonic-clonic seizures
- Concomitant use with valproic acid may produce absence status
Special populations
- Increased production of saliva and bronchial secretion in infants and small children
- Greater pharmacologic effects in elderly patients
- Renal impairment does not affect pharmacokinetics
- Influence of hepatic impairment on pharmacokinetics
Overdose effects
- Symptoms such as drowsiness, ataxia, dysarthria
- Requires monitoring of vital signs and supportive measures
- Prevention of further absorption using activated charcoal
- Severe CNS depression may require flumazenil administration
