Indications

  • Treatment of premature ejaculation in men 18 to 64 years of age
  • Persistent or recurrent ejaculation with minimal sexual stimulation
  • Marked personal distress or interpersonal difficulty as a consequence of premature ejaculation
  • Poor control over ejaculation

Pharmacology

  • Inhibition of neuronal reuptake of serotonin
  • Potentiation of neurotransmitter's action at pre-and post synaptic receptors
  • Rapid absorption with maximum plasma concentrations occurring approximately 1-2 hours after tablet intake
  • Absolute bioavailability of 42%
  • Extensive metabolism to multiple metabolites
  • Rapid distribution with a mean steady state volume of distribution of 162 L
  • Terminal half life of approximately 19 hours

Dosage & Administration

  • Recommended starting dose of 30 mg for adult men (18 to 64 years of age)
  • Maximum recommended dosing frequency of once every 24 hours
  • Dapoxetine may be taken with or without food
  • Not recommended for use in individuals below 18 years of age
  • Caution advised in patients with mild or moderate renal impairment
  • Contraindicated in patients with moderate and severe hepatic impairment

Interaction

  • Caution advised if concomitant administration of Daxetin and CNS active medicinal products is required
  • Daxetin should be prescribed with caution in patients who use PDE5 inhibitors
  • Prescribed with caution in patients who use alpha adrenergic receptor antagonists
  • Caution advised when Daxetin is used in patients taking Warfarin chronically
  • Concomitant use of alcohol and Daxetin could increase the chance or severity of adverse reactions

Contraindications

  • Known hypersensitivity to Dapoxetine Hydrochloride
  • Significant pathological cardiac conditions
  • Concomitant treatment with monoamine oxidase inhibitors (MAOIs) or within 14 days of discontinuing treatment with MAOIs
  • Concomitant treatment with serotonin reuptake inhibitors or within 14 days of discontinuing treatment with these medicinal/herbal products

  • Dry mouth
  • Headache
  • Diarrhea
  • Insomnia
  • Low rates of sexual dysfunction
  • Very mild adverse effects on loss of libido and erectile dysfunction (ED)

Pregnancy & Lactation

  • Not indicated for use by women
  • Unknown excretion through human breast milk

Precautions & Warnings

  • Patients advised not to use Daxetin in combination with recreational drugs
  • Combining alcohol with Daxetin may increase alcohol-related neurocognitive effects
  • Patients could experience syncope at any time during their treatment with Daxetin
  • An orthostatic test should be performed before initiating therapy
  • Caution advised in patients with a history of bleeding or coagulation disorders
  • Pharmacokinetics of Daxetin in combination with Tadalafil and Sildenafil were evaluated in a single dose crossover study

Overdose Effects

  • No case of overdose reported
  • Symptoms of overdose with SSRIs include serotonin-mediated adverse reactions
  • No evidence of withdrawal syndrome in clinical trial
